Wales take on Japan at the Principality Stadium on Saturday, November 7, with kick-off at 4.40pm.
Great Western Railway and Transport for Wales have confirmed they will be running additional trains to Cardiff for the match, as well as for the following fixtures against New Zealand on November 14 and Australia on November 21.
However, all Great Western Railway services ahead of the games will be reservation only, so fans travelling to the capital by train must book their seats in advance.

Rachel Geliamassi, customer services director at Great Western Railway, said: "We’re really pleased to be providing additional trains for this year’s Autumn Internationals at the Principality Stadium.
"Services into Cardiff Central will be reservation only, so we’d urge fans to plan ahead and secure their seats early to avoid disappointment.
"Please allow plenty of time for your journey and remember that trains will be extremely busy both before kick-off and immediately after the matches."
Rail companies warned fans that services will be extremely busy before and after the games, with queuing systems in place to help people board safely.
Cardiff City are also hosting Burnley at the Cardiff City Stadium for a 3pm kick-off, which Transport for Wales said will add further demand on services.
A spokesperson said: "The network will be busier tomorrow with the Wales vs Japan rugby match in the Principality stadium.
"Plan ahead and allow extra time for your journey."
After the match, Transport for Wales customers travelling to the Valleys or Vale of Glamorgan will be directed to Cardiff Queen Street station.
All other queues, including those for Ebbw Vale and Maesteg services, and Great Western Railway trains to Swansea, Bristol and London, will be on the square in front of Cardiff Central station.
An accessibility queue will be available at both stations.
James Gough, customer operations director at Transport for Wales, said: "We’re delighted to be working alongside our partners at Great Western Railway to support the Autumn Internationals and get rugby fans to and from Cardiff safely.
"With extra services running across our networks, we’re encouraging everyone to plan ahead and allow plenty of time for their journey.
"Most importantly, we hope everyone has a brilliant day out and enjoys the game."
Pay-as-you-go is available on Transport for Wales and CrossCountry services across the South Wales Metro network. Passengers should tap in at their departure station and tap out using the same payment method at the dedicated pay-as-you-go lanes when leaving Cardiff Central.
Transport for Wales volunteers will be available to help passengers with the process.
