Three defenders netted for the Lions, with Elliot Owen, Lebrun Mbeka, and Paul Iggulden all scoring after Kallan Murphy's equalising header.
Leek Town had initially taken the lead when Lucas Weir, son of David, headed home in the 23rd minute.
Guiseley began the match with intent, with Amir Berchil and Cam Wilson having early attempts blocked by the Leek Town defence.
Berchil had several efforts in the early stages, but all were thwarted by the away side's sturdy rearguard.
The visitors' goalkeeper, Yusuf Mersin, was kept busy, making a series of saves from the home side's attacking onslaught.
Jack Emmett and Wilson both tested Mersin in the first half, but it was Murphy who eventually found the net with a looping header just before half-time.
The second half started as the first had ended, with Guiseley pushing forward.
Jack Tinker headed over early on before Owen's fantastic finish from a corner gave the home side the lead in the 66th minute.
The Lions never looked back after this, with Mbeka smashing home two minutes later to make it 3-1.
Substitute Iggulden, who replaced the impressive Berchil in the 82nd minute, scored his first goal for the club in the 83rd minute to seal the victory.
The win was Guiseley's eighth in nine games in all competitions, keeping them in third place in the league.
Their next challenge comes in the form of National League side Sutton United, who visit Nethermoor on Saturday, November 7 for an Emirates FA Cup First Round clash.
This match kicks off at 3pm.
