The Championship club confirmed the extent of the 24-year-old's injury almost a week after he was stretchered off during their 4-1 victory over Derby County.
In a statement, QPR said: "The club can confirm Amadou Mbengue will undergo surgery after suffering an ACL injury during our win against Derby last week.
"We are all with you, Amadou."
The update represents a major setback for the former Royals centre-back, who had become a regular figure in QPR's defence following his move from Reading.
Initial fears over the seriousness of the injury were raised immediately after the match when QPR head coach Julien Stephan admitted the signs were concerning.
Speaking after the win over Derby, Stephan said: "We don't know exactly what the situation is with his knee, but it's never good to see a player like this come off with a lot of pain. It's never a good sign."
Mbengue remains a popular figure among Reading supporters after making more than 100 appearances for the club during a challenging period in the Royals' recent history.
The defender joined Reading in 2022 and quickly established himself as a key member of the side before leaving upon the expiry of his contract.
An ACL injury typically requires a lengthy rehabilitation period, meaning Mbengue now faces an extended spell on the sidelines as he begins his recovery.
