The draw took place on Monday evening, just days after Berkhamsted reached the first round for the first time.
While it may not be the blockbuster tie against Luton Town or Leicester City that some would have dreamed of, a match-up against Plymouth Parkway offers the Comrades a chance at progression.
Plymouth Parkway are in the same league as Berkhamsted, although they sit second, nine places above the Comrades.
The two sides have not faced each other so far this season, and their FA Cup tie next month will come well before their first league meeting of the season, in December.
Berkhamsted reached this historic tie after beating Southend United away from home in the final qualifying round at the weekend.
The Comrades prevailed in a historic 2-1 win, courtesy of a 90th-minute Jeffrey Sekyere winner to clinch progression into the first round.
First-round ties are set to be played across the weekend of Saturday, November 7, with confirmation of exact dates and times to follow in due course, once broadcast selections take place.
