Sweeeney drew Redditch level after Tyler Bruck put the Reds ahead only for Harry Williams to score a brace for Gloucester despite the visitors playing with 10-men after Michael O’Regan saw red.
However, the Reds scored all four of their penalties in the shootout to reach the second round while Gloucester failed to hit the target with two of their spot kicks from 12-yards.
Redditch broke the deadlock in an uneventful first half on 41 minutes as Bruck controlled Szhem Whyte-Hall’s cross and coolly found the bottom right corner from 12-yards.
And Southern League Premier South side Gloucester then had O’Regan sent off five minutes after the break after he tripped Bruck as the last defender.
However, the 10-men equalised on 63 minutes when Williams finished from close range after Rory Brown failed to punch a corner from the left to safety.
And Williams completed the turnaround seven minutes later when he turned home a low ball from the right at the near post with a finish across goal into the bottom left corner.
But the Reds forced a shootout two minutes from time when sub Sweeney brought down a long throw from the right and finished on the turn from six-yards out.
The Reds stayed perfect from the spot after four spot kicks as Danny King, Sweeney, Bruck and Harrison Hazard all converted from 12-yards.
However, Ethan Dunbar blazed over and Kian Hill shot wide for Gloucester to send United into the next round of the competition.
Redditch are next in action against Racing Club Warwick on Tuesday, October 27 with kick-off at 7.45pm at Townsend Meadow.
